 Effects of auxin on seed germination

1, Principle

Auxin and its synthetic analogens, such as naphthylacetic acid, have great regulatory effects on plant growth, and their effects on plant growth are also different at different concentrations. In general, low concentrations of auxin promote growth and high concentrations inhibit it. Different plant organs respond differently to auxin, with roots generally more sensitive to auxin than buds and stems. The effects of different concentrations of naphthalene acetic acid on the growth of different plant organs during seed germination were observed.

 

2.Materials, instruments, equipment and reagents

(1) Materials: wheat seeds.

(2) Equipment: temperature box, petri dish, pipette, tweezers, filter paper, ruler.

Reagent: 10mg/L naphthalene acetic acid, 0.1% mercury.

 

3. Experimental steps

1. Wheat seeds were taken, disinfected with 0.1% hg for 15min, then washed with tap water and distilled water for 3 times respectively, and placed in a temperature box at 22℃ to accelerate germination for 2d.

2. 7 sets of clean 9cm Petri Dishes were taken and numbered. Add 10mg/L naphthalene acetic acid 10mL to petri dish no. 1, then absorb 1mL of it into Petri dish No. 2, add 9mL distilled water and mix to form 1 mg/L naphthalene acetic acid solution. In this way, the concentration of naphthylacetic acid in No.1 to 6 petri dishes was 10 mg/L, 1 mg/L, 0.1 mg/L, 0.01 mg/L, 0.001 mg/L and 0.0001 mg/L, respectively, after being successively diluted to No. 6 petri dishes (1mL was finally removed from No. 6 Petri dishes). In petri dish No. 7, 9mL distilled water was added as control.

3. Put a filter paper with the same size as the bottom of the petri dish into each petri dish, select 10 germinating wheat seeds, arrange them neatly in the petri dish with tweezers, so that the bud tip is upward and the part of the embryo is toward the same side, cover the dish lid, and put it in a temperature box at 22℃ for dark culture for 3 days.

4. Measure the average length of wheat buds and young roots and the number of roots in the culture dish.
