Inoculation loop operation steps
The prepared plate can be stored for 2-5 days when the inoculation loop is stored, and direct light exposure should be avoided. The dry medium should be placed in a dark and dry place, and the storage temperature should be 2-8°C. Expired dry media should not be used beyond their shelf life, clumping and color changes.
Steps:
1. Use an inoculation loop to inoculate 10 μl of broth or decoction to streak or spread it on a color plate of Enterobacter sakazakii;
2. Put it upside down in an incubator at 36±1℃ for 22-26h, the typical Enterobacter sakazakii is a blue-green colony;
3. Pick typical colonies for biochemical tests, which have good fluidity and are pale yellow or milky white. The prepared plate is pale yellow solid medium.
