Isolation of chloroplast pigments
(1) Take a piece of circular qualitative filter paper (11cm in diameter) and poke a small circular hole (about 3mm in diameter) in its center. Take another filter paper strip (5cm×1.5cm), use a dropper to draw the ethanol chloroplast pigment extract and apply it to one side of the paper strip along the length direction of the paper strip, so that the width of the pigment diffusion is limited within 0.5cm, and after air-drying, Repeat the operation several times, and then roll it into a paper twist along the length direction, so that the side soaked in the chloroplast pigment solution is just at one end of the paper twist.
(2) Insert the pigmented end of the paper twist into the small hole of the circular filter paper so that it is just flush with the filter paper (do not protrude).
(3) A short flat-bottomed glass tube or a plastic medicine bottle cap can also be placed in the petri dish to hold the propellant. The diameter of the bottom and lid of the petri dish used should be the same and slightly smaller than the diameter of the filter paper. Place the circular filter paper with the paper twist on the petri dish and immerse the lower end of the paper twist in the propellant. Quickly cover the petri dish. At this time, the propellant spreads to the circular filter paper along the paper twist by capillary gravitational force, and pushes the chloroplast pigments around, and soon concentric rings of various pigments can be seen.
(4) When the front of the propellant is close to the edge of the filter paper, take out the filter paper and air-dry, you can see the various pigments separated: chlorophyll a is blue-green, chlorophyll b is yellow-green, lutein is bright yellow, and carotene is orange yellow. Use a pencil to mark the locations and names of the various pigments.
