Experiment of slant medium inoculation method
This method is suitable for pure culture and preservation of strains, and can also be used for the identification test of urea and Krebsaccharide medium. The method of inoculation is slightly different due to different purposes. The slant inoculation method for pure culture is to sterilize the inoculation loop (needle), pick a single colony and draw a straight line from the bottom of the medium, and then inoculate it on the slant of the agar with a snake-shaped streak. Use the inoculation needle to pick the bacteria to be identified. Generally, the top of the colony should be taken. First, insert the inoculation needle into the middle of the inclined plane and pierce the bottom vertically. After pulling it out, make a serpentine line on the inclined plane.
