How microorganisms are inoculated on a slant

(1) On the slanted test tube of meat extract peptone, use a marker to write the name of the bacteria to be inoculated, the date and the inoculated person.

(2) Light an alcohol lamp or a gas lamp.

(3) Hold the strain test tube and the inclined test tube to be inoculated in the left hand with the thumb, index finger, middle finger, and ring finger, and sandwich the middle finger between the two test tubes, so that the inclined surface is upward and in a horizontal state. Loosen the stopper of the test tube with your right hand beside the flame to facilitate extraction during inoculation.

(4) Hold the inoculation ring in the right hand and sterilize it by flame cauterization. Hold the cotton plug (or test tube cap) with the palm edge of the right hand and the little finger, the little finger and the ring finger respectively at the flame, take it out, and quickly cauterize the mouth of the tube.

(5) Extend the sterilized inoculation loop into the bacterial culture test tube, first touch the loop to the inner wall of the test tube or the culture medium without bacteria to achieve the purpose of cooling, and then pick a little bacterial moss. Take the inoculation loop out of the strain test tube, quickly extend it into the inclined test tube to be inoculated, and use the loop to gently draw a straight line from the bottom to the top of the test tube on the inclined surface. Do not cut the medium, and do not let the loop touch the tube wall or mouth .

(6) The inoculation ring exits the inclined test tube, then burn the mouth of the tube with a flame, and plug the test tube on the flame. Gradually bring the inoculation loop close to the flame and then cauterize. If there are too many bacteria on the inoculation ring, the ring should be dried by the flame first, and then burned, so as to prevent the unburned bacteria from splashing out and polluting the environment. Pay more attention to this point when inoculating pathogenic bacteria.
