Method for separating actinomycetes from soil with petri dish

1. Make Gao’s No. 1 medium, pour it into a petri dish while it is hot, and condense it into a flat plate, set aside.High Quality Petri Dish For Lab Use,Petri Dish Plastic Disposable,Plastic Petri Dish
2. Weigh 10 grams of soil, put it into an Erlenmeyer flask containing 100 ml of sterile water, and add 10 drops of 10% phenol to inhibit bacterial growth. Shake for 10 minutes to make a 10-1 bacterial suspension. According to the serial dilution separation method, a 10-3 bacterial suspension is further prepared.
3. Use a transfer pipette to draw 0.1 ml of 10-3 bacterial suspension, pour it into the plate medium, and use a sterile glass spatula to evenly spread the bacterial suspension on the entire medium. Then put the petri dish upside down in an incubator at 25-30°C and cultivate for 7-10 days.High Quality Petri Dish For Lab Use,Petri Dish Plastic Disposable,Plastic Petri Dish Microbial colonies will appear on the culture medium. If the colony is hard, dry and compact, and tightly combined with the substrate, it is not easy to be picked up by the needle, this is the actinomycete colony.
4. Pick the colonies of actinomycetes and inoculate them on the slant medium.
