Sampling: In food processing plants, the number of samples taken is per shift. If the finished product is less than 5t, sample one can; 5~10t sample two cans. Preparation of sample solution: Tomato juice and sauce can be directly sampled. Tomato paste or tomato paste must be diluted with water to form a sample solution with a solid content equivalent to a refractive index of 1.3447~1.3460 at 20 °C, which is a standard sample solution (refractive index using Abbe). refractometer measurement).

Standard preparation: Weigh 10g (concentration of about 28.5%) tomato paste on a tray balance with a small burning cup, add 26ml of distilled water, and stir evenly with a glass rod, that is, the standard sample solution with a refractive index of 1.3447~1.3460 (or with sugar content) The standard sample solution with a concentration of 7.9% to 8.8% was determined by the meter) for use. If the refractive index is too large or too small, water or sample solution must be added until the standard sample solution is prepared before testing.

4. Smear
(1) First, wipe the microscope slide or microscope cover glass with lens cleaning paper or cloth and alcohol until the microscope cover glass is changed to produce Newton’s rings on the two shoulders of the microscope slide.
(2) Take a large drop of the well-mixed standard sample solution with a dropper or glass rod, spread it evenly on the flat surface in the center of the microscope slide, and cover it with a microscope cover glass. If it is found that the sample solution is unevenly coated, there are air bubbles, or the sample solution flows into the groove and flows out from the microscope cover glass and the shoulder bank, that is, the shoulder bank of the microscope cover glass and microscope slide does not produce Newton rings, it should be discarded.
