[Principle]
The root system of a plant is an organ that is extremely active in life activities. It can synthesize some substances necessary for life, supply other organs, and at the same time excrete some substances from the body, changing the surrounding environment (soil), thereby affecting the growth of other organisms. Here we only observe the common exudates of plant roots.
[Steps]
1. Amino acid detection
The wheat or corn seeds are germinated in a petri dish, and when the roots are 2-3cm long, they can be used as test materials. Carefully remove the germinated seeds and place them in another petri dish with damp filter paper, and cover with a piece of damp filter paper. In order to make the root system contact with the filter paper well, the embryo can also be removed, and the petri dish is placed in the incubator overnight. Remove the seeds, dry the filter paper, spray the ninhydrin alcohol solution, and bake it in an oven at 80°C for 1 quarter of an hour. A blue-purple color can be seen where the filter paper has touched the root system, indicating the presence of amino acids.
2. Sugar detection
First prepare the germinated wheat or corn, and the test can be carried out when the root system is 3─5cm. Take several seedlings and place them in a small test tube. Be careful not to damage the root system. Pour 5×10-4 mol/L CaSO4 solution into the test tube to immerse the root system as much as possible (as little as possible) and place it in the incubator overnight. Then use a pipette to suck 2ml of the solution into a test tube, add 5ml of the anthrone reagent, and heat it in a boiling water bath for 10 minutes. If the solution turns green, it indicates the presence of sugars.
3. Detection of amylase
Pour the prepared 0.02% starch agar (2%) solution into a petri dish to make a thin plate, then take the germinated seeds and place them on the agar plate, so that the roots are in contact with the agar as much as possible, and placed in the incubator Several hours or overnight. Then remove the seeds, pour a small amount of dilute I-KI solution on the plate, you can see a transparent starch-free area where the root system has grown.
