Pipetting techniques for special samples

In most cases, the pipetting object of the pipette is aqueous solution, but it is inevitable to encounter some special samples, such as high volatility, high viscosity, high density or very warm samples. If you need to transfer these special samples frequently, we suggest you to buy a pipette with external piston principle. Although the cost of consumable materials is a little high, the pipetting accuracy is guaranteed after all, isn’t it? If you only transfer special samples occasionally and have limited financial resources, please follow me below.

Pipetting techniques for special samples
1. Two points should be paid attention to when removing volatile samples:
A. Be sure to rinse twice before pipetting;
B. Drain after suction is complete.
2 remove the sample with high viscosity
Reverse pipetting mode is adopted: push the suction/discharge button to second gear (second stop point) for suction and push the button to second gear (first stop point) for drainage. In addition, the retention time of 3-5 seconds is required for both suction and drainage.

3. Remove high/low density samples
The pipette accuracy is based on transferring pure water. If the density of the sample differs greatly from that of the water, the accuracy will also differ greatly. Therefore, it is necessary to find out the density of the sample before pipetting, and then adjust the range to the product of the sample volume and density to be transferred.
For example, if a sample density is 1.2g/cm3 and you need to transfer 300ul, you should set the range to 360ul. This is only a rough adjustment method, strict adjustment method also needs to use a gauge or balance as an auxiliary tool for accurate calculation.
4. Remove high/low temperature samples
Three points should be paid attention to when removing high/low temperature samples:
A. Do not wash the suction head before pipetting;
B. A new pipette should be replaced each time;
C. Suction and drainage should be completed as soon as possible.
