Agar slant medium
Isolated cultured plate cultures.
(2) Method
①Indicate the name (or serial number) of the bacteria to be transplanted, the date of inoculation, the group and code of the inoculated person on the upper part of the test tube of the agar slant medium.
②The left hand holds the bottom of the plate with bacteria, and the right hand holds the inoculation loop. After the inoculation loop is sterilized and cooled on a flame, the well-developed isolated colonies on the streak on the plate are dipped. Put the plate back to its original place, immediately take the slanted medium with this hand and hold it obliquely, hold the cotton plug with the little finger and the palm of the right hand, turn the cotton plug gently and remove the cotton plug, and the nozzle is sterilized by flame.
③ Extend the infected inoculation loop into the solidified water at the bottom of the tube of the slant medium, draw a straight line from the bottom to the top along the surface of the slant medium, and then draw a continuous zigzag line from the bottom up; it is also possible to draw a zigzag line without a straight line . Take out the inoculation loop, sterilize the tube mouth by flame, plug the cotton plug, put it on the test tube rack, and put the inoculation loop down after flame sterilization.
If the bacteria transplanted to the slant medium are grown on the slant medium or in the liquid medium, use the left hand to hold the two tubes of the seed tube and the slant medium at the same time, and the ring finger and little finger of the right hand. Pull out and hold two tampons at the same time with the palm of your hand. After the nozzle and the inoculation ring are first sterilized by flame, the bacteria are taken from the strain tube and immediately transferred to the inclined surface, the cotton plug is plugged, and the inoculation ring is put down after flame sterilization. Then the slant medium was placed in a 37°C incubator for 24h. As a result, bacterial moss was formed on the inclined surface.
