The effect of chemical factors on bacteria (sterilization test of chemical disinfectants)

【Material】
1. Bacteria: Staphylococcus agar slant culture for 18-24 hours.
2. Medium: ordinary agar plate.
3. Chemical disinfectant: 5% carbolic acid, 2% iodine, 70% alcohol, 0.1% mercury chloride.
4. Others: sterile round filter paper with a diameter of 0.6cm, tweezers, inoculation loop, etc.
【Method】
1. Divide the agar plate into quarters and mark the bottom.
2. Use an inoculating loop to take a slanted culture of Staphylococcus agar and spread it over the entire surface of the agar medium.
3. Use small tweezers to pick up the sterile small filter paper, and dip them in the above disinfectant respectively (do not dip too much disinfectant to prevent outflow). It is flatly attached to the center of each corresponding partition, covered with a lid, and the date and tester are marked.
4. After incubating at 37℃ for 18~24 hours, observe the bactericidal effect of various chemical disinfectants. The area without bacterial growth around the paper sheet is called the inhibition zone, and the diameter of the inhibition zone of the four disinfectants is measured and recorded in millimeters.
