[Culture in liquid medium]
⑴ Take 5ml of liquid culture medium into a sterile test tube.
(2) Pick a single colony with an inoculation loop or a sterilized toothpick and inoculate it into the culture medium.
(3) Cover the test tube and incubate overnight at 37°C on a shaker at a speed of 60 r/min.

[Cultivation in solid medium]
Bacteria are grown on solid media mainly for single colonies and short-term storage.
(1) Using aseptic technique, use an inoculation loop to streak the inoculum from one side of the culture plate.
(2) Re-sterilize the inoculation loop, streak the sample from the first streak to the rest of the culture plate, and repeat the streak until the entire culture plate is covered.
(3) Incubate at 37°C until a single colony grows.
